RESUMO

Environmental stresses diversely affect multiple processes related to the growth, development, and yield of many crops worldwide. In response, plants have developed numerous sophisticated defense mechanisms at the cellular and subcellular levels to react and adapt to biotic and abiotic stressors. RNA silencing, which is an innate immune mechanism, mediates sequence-specific gene expression regulation in higher eukaryotes. ARGONAUTE (AGO) proteins are essential components of the RNA-induced silencing complex (RISC). They bind to small noncoding RNAs (sRNAs) and target complementary RNAs, causing translational repression or triggering endonucleolytic cleavage pathways. In this review, we aim to illustrate the recently published molecular functions, regulatory mechanisms, and biological roles of AGO family proteins in model plants and cash crops, especially in the defense against diverse biotic and abiotic stresses, which could be helpful in crop improvement and stress tolerance in various plants.

BACKGROUND: Oogenesis is a complex pathway necessary for proper female reproduction in insects. Ovary-serine protease (Osp) is a homologous gene of serine protease Nudel (SpNudel) and plays an essential role in the oogenesis and ovary development of Drosophila melanogaster. However, the function of Osp is not determined in Plutella xylostella, a highly destructive pest of cruciferous crops. RESULTS: The PxOsp gene comprises a 5883-bp open-reading frame that encodes a protein consisting of 1994 amino acids, which contain four conserved domains. PxOsp exhibited a high relative expression in adult females with a specific expression in the ovary. Through the utilization of CRISPR/Cas9 technology, homozygous mutants of PxOsp were generated. These homozygous mutant females produced fewer eggs (average of 56 eggs/female) than wild-type (WT) females (average of 97 eggs/female) when crossed with WT males, and these eggs failed to hatch. Conversely, mutant males produced normal progeny when crossed with WT females. The ovarioles in homozygous mutant females were significantly shorter (5.02 mm in length) and contained fewer eggs (average of 3 eggs/ovariole) than WT ovarioles (8.09 mm in length with an average of 8 eggs/ovariole). Moreover, eggs laid by homozygous mutant females were fragile, with irregular shapes, and were unable to maintain structural integrity due to eggshell ruptures. However, no significant differences were observed between WT and mutant individuals regarding developmental duration, pupal weight, and mating behavior. CONCLUSION: Our study suggesteds that PxOsp plays a vital role in female reproduction, particularly in ovary and egg development. Disrupting PxOsp results in recessive female sterility while leaving the male reproductive capability unaffected. This report represents the first study of a haplosufficient gene responsible for female fertility in lepidopteran insects. Additionally, these findings emphasize PxOsp as a potential target for genetically-based pest management of P. xylostella. © 2024 Society of Chemical Industry.

Cadmium (Cd) contamination is an eminent dilemma that jeopardizes global food safety and security, especially through its phytotoxicity in rice; one of the most edible crops. Melatonin (MET) has emerged as a protective phytohormone in stress conditions, but the defensive role and underlying mechanisms of MET against Cd toxicity in rice still remain unclear. To fulfill this knowledge gap, the present study is to uncover the key mechanisms for MET-mediated Cd-stress tolerance in rice. Cd toxicity significantly reduced growth by hindering the process of photosynthesis, cellular redox homeostasis, phytohormonal imbalance, and ultrastructural damages. Contrarily, MET supplementation considerably improved growth attributes, photosynthetic efficiency, and cellular ultrastructure as measured by gas exchange elements, chlorophyll content, reduced Cd accumulation, and ultrastructural analysis via transmission electron microscopy (TEM). MET treatment significantly reduced Cd accumulation (39.25%/31.58%), MDA (25.87%/19.45%), H2O2 (17.93%/9.56%), and O2 (29.11%/27.14%) levels in shoot/root tissues, respectively, when compared with Cd treatment. More importantly, MET manifested association with stress responsive phytohormones (ABA and IAA) and boosted the defense mechanisms of plant by enhancing the activities of ROS-scavenging antioxidant enzymes (SOD; superoxide dismutase, POD; peroxidase, CAT; catalase, APX; ascorbate peroxidase) and as well as regulating the key stress-responsive genes (OsSOD1, OsPOD1, OsCAT2, OsAPX1), thereby reinstate cellular membrane integrity and confer tolerance to ultrastructural damages under Cd-induced phytotoxicity. Overall, our findings emphasized the potential of MET as a long-term and cost-effective approach to Cd remediation in paddy soils, which can pave the way for a healthier and more environmentally conscious agricultural sector.

BACKGROUND: Cysts of the jaws constitute an important pathology in the oral and maxillofacial region and are broadly classified as odontogenic and non-odontogenic. Objective of this study was to document the clinical and radiographic presentation of odontogenic cysts at a tertiary care centre. METHODS: In this descriptive case-series, patients presenting with features of suspected cystic lesions of the jaws were investigated using radiographs and incisional biopsy. Subjects showing clinico- radiographic features of odontogenic cyst(s) with subsequent confirmation on histopathological examination were included. RESULTS: A total of 112 subjects were investigated for suspected jaw cysts and cystic diagnosis of odontogenic cysts was confirmed in 100 patients with 53 males (53%) and 47 females (47%). The age range of patients was from 12-55 years. Radicular cyst was the most commonly diagnosed odontogenic cyst (63%) followed by dentigerous cyst (22%) and odontogenickeratocyst (14%). Anterior maxilla was the most affected site (44%) followed by posterior mandible (30%). CONCLUSION: Odontogenic cysts constitute an established pathology affecting the jaws with varying frequency. This study augments the data cited from Western countries and re-emphasizes the need for early diagnosis and prompt management. It also highlights the differences in the presentation of odontogenic cysts as observed in the current series.
